摘要: The effects of eicosapentaenoic acid on synthesis and secretion cholesterol ester by cultured rat hepatocytes were studied. In the presence cellular esterification was decreased 50-75% compared to oleic as measured radioactive precursors mass. Secretion reduced 50-60% in evaluated radiolabeled fatty acids, mevalonolactone, mass measurement. Oleic, palmitic, stearic increased, whereas docosahexaenoic a acid-free control. Cellular secreted free unaffected comparison with acid. observed within 1 h lasted for at least 20 h. Eicosapentaenoic caused lower than concentration range 0.2-1.0 mM stimulatory effect formation. Cholesterol release markedly increased 25-hydroxycholesterol well Experiments liver microsomes revealed that eicosapentaenoyl-CoA poorer substrates (7-30%) oleoyl-CoA. Reduced formation also when given together labeled oleoyl-CoA, palmitoyl-CoA, stearoyl-CoA, linolenoyl-CoA, arachidonoyl-CoA had no inhibitory effect. conclusion, inhibiting activity acyl-CoA:cholesterol acyltransferase. lowered secondly very low density lipoprotein ester.
