摘要: The gene encoding rat somatostatin has been isolated from a lambda phage library. Phage harboring the were identified by plaque hybridization using nick-translated fragment derived cDNA for somatostatin. transcriptional unit includes exons of 238 and 367 base pairs (bp) separated one intron 621 bp. is located between codons Gln (-57) Glu (-56) prosomatostatin. Analysis nucleotide sequence 5' to start transcription reveals number sequences which may be involved in expression A variant "TATA" box, TTTAAA, lies 26 bp upstream transcription, homologous "CAAT" box (GGCTAAT) 92 start. long alternating purine-pyrimidine stretch, (GT)25, similar Z DNA-forming other genes, 628 flanked small repeats. Hybridization analysis shows that this region highly repeated genome are approximately 2 kilobase downstream poly(A) addition site. Southern clone with probes brain or liver poly(A+) RNA demonstrates another transcribed about 7 site gene. DNA suggests there restriction-site polymorphisms near additional somatostatin-hybridizing exist genome.
