摘要: We have evaluated the possible role of proteoglycans in uptake spermine by human lung fibroblasts. Exogenous glycosaminoglycans behaved as competitive inhibitors uptake, most efficient being heparan sulphate (Ki=0.16+/-0.04 microM). Treatment fibroblasts with either lyase, p-nitrophenyl-O-beta-D-xylopyranoside or chlorate reduced considerably, whereas chondroitin lyase had a limited effect. Inhibition polyamine biosynthesis alpha-difluoromethylornithine resulted an increase cell-associated exhibiting higher affinity for spermine. The data indicate specific Spermine pgsD-677, mutant Chinese hamster ovary cell defective biosynthesis, was only moderately (20%) compared wild-type cells. cells above-mentioned xyloside greater reduction endogenous proteoglycan production well inhibition than Moreover, treatment selective cells, indicating chondroitin/dermatan these Fibroblasts, made growth-dependent on exogenous treatment, were growth-inhibited beta-D-xyloside, which might future therapeutical implications.
