Analysis of storage proteins in normal and aborted seeds from embryo-lethal mutants of Arabidopsis thaliana.

摘要: The major storage proteins isolated from wild-type seeds of Arabidopsis thaliana (L.) Heynh., strain “Columbia”, were studied by sucrose gradient centrifugation and sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE). Both the hypocotyl cotyledons mature embryos contained abundant 12 S (cruciferin) 2 (arabin) that appeared similar in size subunit composition to cruciferin (12 S) napin (1.7 seed-storage Brassica napus. protein was resolved SDS-PAGE into two groups subunits with approximate relative molecular weights 22–23 kDa (kilodalton) 30–34 kDa. These polypeptides accumulated late embryo development, disappeared early germination, not detected other vegetative or reproductive tissues. Accumulation aborted nine embryo-lethal mutants different patterns abnormal development determine extent cellular differentiation arrested each mutant line. Abundant found lethal phases, but seven phases ranging globular cotyledon stages development. results indicate accumulation is closely tied morphogenetic changes occur during Embryo-lethal may therefore be useful future studies on developmental regulation storage-protein synthesis.
