Are minichromosomes valid model systems for DNA replication control? Lessons learned from Escherichia coli

摘要: Initiation of chromosome replication is a key event in the life cycle any organism. Little known, however, about regulatory mechanisms this vital process. Conventionally, initiation mechanism microorganisms has been studied using plasmids which an origin cloned, rather than itself. The reason for that even bacterial chromosomes are so large biochemical and genetic manipulations become difficult cumbersome. Recently, combination flow cytometry methods, modifications systematically introduced onto chromosome, made possible detailed studies molecular events involved control Escherichia coli. results indicate requirements at chromosomal origin, oriC, drastically different from those cloned oriC.
