摘要: Phagocytosis of apoptotic cells (efferocytosis) induces macrophage differentiation towards a regulatory phenotype (IL-10high/IL-12p40low). CD36 is involved in the recognition (AC), and we have shown that platelet-activating factor receptor (PAFR) also involved. Here, investigated contribution PAFR to efferocytosis establishment phenotype. Mice bone marrow-derived macrophages were cocultured with thymocytes, phagocytic index was determined. Blockage antagonists or specific antibodies inhibited phagocytosis AC (~70–80%). Using immunoprecipitation confocal microscopy, showed increased colocalisation plasma membrane; coimmunoprecipitated flotillin-1, constitutive lipid raft protein, disruption these membrane microdomains by methyl-β-cyclodextrin reduced phagocytosis. Efferocytosis induced pattern cytokine production, IL-10high/IL-12p40low, is, characteristic LPS potentiated efferocytosis-induced production IL-10, this prevented blocking CD36.
