摘要: Whereas previously we have successfully utilized the folding funnels concept to rationalize binding mechanisms (Ma B, Kumar S, Tsai CJ, Nussinov R, 1999, Protein Eng 12:713-720) and describe (Tsai Ma Sci 8:1181-1190), here further extend of funnels, illustrating its utility in explaining enzyme pathways, multimolecular associations, allostery. This extension is based on recognition that are not stationary; rather, they dynamic, depending physical or conditions Proc Natl Acad USA 96:9970-9972). Different states change surrounding environment proteins. The changed turn expressed shifted energy landscapes, with different shapes distributions populations conformers. Hence, function a protein properties only decided by static folded three-dimensional structure; determined distribution conformational substates, particular, redistributions under environments. That is, derives from dynamic landscape, caused changes surroundings.
