A chimeric human-cat fusion protein blocks cat-induced allergy

摘要: Animal allergens are an important cause of asthma and allergic rhinitis. We designed tested a chimeric human-cat fusion protein composed truncated human IgG Fcgamma1 the major cat allergen Fel d1, as proof concept for new approach to allergy immunotherapy. This Fcgamma-Fel d1 induced dose-dependent inhibition d1-driven IgE-mediated histamine release from cat-allergic donors' basophils sensitized cord blood-derived mast cells. Such was associated with altered Syk ERK signaling. The also blocked in vivo reactivity FcepsilonRIalpha transgenic mice passively IgE antibody Balb/c actively against d1. alone did not induce mediator release. Chimeric Fcgamma-allergen proteins may provide therapeutic platform immune-based therapy disease.
