摘要: Enriched CD34(+) peripheral blood progenitor cells (PBPC) are frequently used as stem cell support in cancer patients following high dose therapy. Since precursor dendritic (DCs) originate from haematopoietic cells, purified might also serve starting for ex- vivo production of DC. In the present study we developed a clinical grade procedure DC derived enriched cells. Different concentrations were grown gas-permeable Teflon bags with different serum-free and serum-containing media supplemented GM-CSF, IL-4, TNF-alpha, SCF, Flt-3L INF-alpha. Serum-free CellGroSCGM medium 7 days followed by CellGroDC gave same results medium. After incubation cultured containing immature DCs concentrated transfected tumour mRNA human prostate lines employing highly efficient electroporation procedure. Thawed able to elicit primary T-cell responses vitro against antigens encoded shown ELISPOT assay using mock-transfected control. Our show that frozen can be an alternative source therapeutic purpose.
