摘要: Stability of the human alpha-globin mRNA is conferred by a ribonucleoprotein complex termed alpha-complex, which acts impeding deadenylation. Using our recently devised in vitro decay assay, we demonstrate that alpha-complex also functions protecting 3'-untranslated region (3'-UTR) from an erythroid-enriched, sequence-specific endoribonuclease activity. The cleavage site was mapped to protected and regulated presence alpha-complex. Similar products were detected erythroid cells expressing exogenous gene. Nucleotide substitution target sequence renders RNA refractory Insertion onto heterologous confers on chimeric RNA, demonstrating specificity this We conclude stabilizes multifaceted approach, one aspect protect 3'-UTR specific cleavage.
