CLEC4F, A Kupffer Cells Specific Marker, Is Critical for Presentation of Alfa-Galactoceromide to NKT Cells (78.38)

摘要: CLEC4F, a member of C-type lectins, was firstly purified from rat liver extract with high binding affinity to fucose, galactose and N-acetylgalactosamine, un-sialylated glucosphingolipids GalNAc or Gal terminus. However, the biological functions CLEC4F have not been elucidated. To address this question, we generated mAbs detect expression murine (mCLEC4F) in vivo, investigated its function using mCLEC4F-deficient mice. The anti-mCLEC4F bind 293T cells transfected mCLEC4F cDNA as determined by flow cytometry, specificity is further confirmed Histochemical staining showed that only expressed on F4/80+ localized liver, undetectable bone marrow, spleen, lymph nodes, other tissues adult detected embryonic day 11.5 (E11.5), which 1.5 earlier than formation (E10) 3.5 marrow (E15-16). Moreover, recombinant mCLEC4F.Fc binds alpha-galactoceramide Ca++-dependent manner, both ceramide can partially inhibit CLEC4F.Fc alpha-galactoceramide. Interestingly, (mCEC4F k/o) mice produced far less cytokines wild type littermates after intravenous injection ofalpha-galactoceramide. This suggests specific marker for Kupffer cells, but also critical presentation glycolipid antigen NKT cells.
