摘要: The success of gene therapy depends on efficient delivery DNA and requires a vector. A promising non-viral vector is chitosan. We tailored chitosan to optimize it for transfection by synthesizing self-branched trisaccharide-substituted oligomers (SBTCO), which show superior efficacy compared with linear (LCO). aim the work was compare cellular uptake endocytic pathways polyplexes formed LCO SBTCO. Both were taken up majority cells, but lower than SBTCO polyplexes. internalized through both clathrin-dependent clathrin-independent pathways, whereas primarily endocytosis. different level distinct may explain difference in efficacy.
