Activation of Peroxisome Proliferator-Activated Receptors in Human Airway Smooth Muscle Cells Has a Superior Anti-inflammatory Profile to Corticosteroids: Relevance for Chronic Obstructive Pulmonary Disease Therapy

摘要: Airway smooth muscle is actively involved in the inflammatory process diseases such as chronic obstructive pulmonary disease and asthma by 1) contributing to airway narrowing through hyperplasia hypertrophy 2) release of GM-CSF G-CSF, which promotes survival activation infiltrating leukocytes. Thus, identification novel anti-inflammatory pathways will have important implications for treatment disease. This study identifies a pathway peroxisome proliferator-activated receptors (PPARs). PPAR ligands are known therapeutic agents diabetes; however, their role human unknown. We demonstrate, first time, that cells express PPARα -γ subtypes. Activation PPARγ natural synthetic inhibits serum-induced cell growth more effectively than does steroid dexamethasone, induces apoptosis. Moreover, activation, like GM-CSF. However, ligands, but not similarly G-CSF release. These results reveal muscle, where has additional effects those steroids. Hence, might act potential treatments respiratory diseases.
