摘要: Human pluripotent stem cells (hPSCs), including embryonic and induced cells, provide a powerful platform for mechanistic studies of disorders neurodevelopment neural networks. hPSC models autism, epilepsy, other neurological are also advancing the path toward designing testing precision therapies. The field is evolving rapidly with addition genome-editing approaches, expanding protocols two-dimensional (2D) differentiation different neuronal subtypes, three-dimensional (3D) human brain organoid cultures. However, application these techniques to study complex disorders, epilepsies, remains challenge. Here, we review previous work using both 2D 3D genetic as well recent advances in field. We describe new strategies applying technologies disease modeling discuss current challenges future directions.
