摘要: Glucocorticoids rapidly and specifically inhibit transcription of the pro-opiomelanocortin (POMC) gene in anterior pituitary, thus offering a model for studying negative control mammals. We have defined an element within rat POMC 5'-flanking region that is required glucocorticoid inhibition POMC-expressing pituitary tumor cells (AtT-20). This contains vitro binding site purified receptor. Site-directed mutagenesis revealed receptor to this located at position base pair -63 essential repression transcription. Although related well-defined response (GRE) found glucocorticoid-inducible genes, DNA sequence (nGRE) differs significantly from GRE consensus; divergence may result different receptor-DNA interactions account least part opposite transcriptional properties these elements. Hormone-dependent be due over positive regulatory promoter. Thus, mutually exclusive two DNA-binding proteins overlapping sequences.
