摘要: Metastatic spread depends critically upon the invasiveness of tumor cells, i.e. their ability to breach basement membranes by elaborating and secreting specific proteolytic enzymes such as gelatinase A (MMP-2). Laminin is a major constituent extracellular matrix that can trigger production MMP-2 in metastatic but not non-metastatic cells. The present study was designed examine role phospholipase D (PLD) its product, phosphatidic acid, intracellular signal transduction mechanisms mediate induction laminin. Here we show stimulation cells with laminin results time- dose-dependent activation PLD. Laminin-induced attenuated 1-butanol, competitive substrate PLD reduces PLD-catalyzed PA. Moreover, acid itself induce also be induced exposing exogenous bacterial Elevated cellular induces ras-transformed 3T3 fibroblasts but, like laminin, fails do so normal These data indicate laminin-induced consequent generation are involved propagation pathway leading enhanced
