摘要: The aim of the present study was to determine whether pharmaceutical preparations with pregabalin (PGB) as an active ingredient, which are widely prescribed by clinicians, exert toxic effects on human primary nucleus pulposus (NP) and annulus fibrosis (AF). Primary cell cultures were obtained from intact (n=6) degenerated tissues resected two groups patients. Different doses PGB applied these cells subjected molecular analyses at 0, 24 48 h. Cell vitality, toxicity proliferation assessed using a spectrophotometer. expression chondroadherin (CHAD), (member NP-specific protein family), hypoxia-inducible factor-1α (HIF-1α) type II collagen (COL2A1) measured reverse transcription-quantitative polymerase chain reaction. results revealed that intensity increased in time-dependent manner vitality continued without pharmaceuticals. suppressed PGB-treated independent dose duration application. demonstrated suppress CHAD HIF-1α. In contrast, COL2A1 gene not any experimental group. utilized vitro model ingredient used herein may be representative clinical applications; however, has effect NP/AF containing intervertebral disc tissue. summary, use pharmacological agents differentiation and/or tissues, should considered when deciding appropriate treatment regime.
