摘要: Signal transducer and activator of transcription protein 3 (STAT3) is a latent cytosolic factor that widely recognized as being master regulator the cellular functions lead to cancer phenotype. Constitutively activated STAT3 activity routinely observed in human cancers, promoting uncontrolled cell proliferation suppressing apoptosis. Until relatively recently, inhibition transcriptional was achieved indirectly via suppression upstream kinase activators extracellular cytokine (or) growth stimuli. However, forms transcriptionally functional STAT3-STAT3 dimers, providing valid juncture for targeted downstream molecular inhibition. STAT3's prominent role has seen decade innovative novel approaches targeting constitutively active protein-protein complexes. This mini-review outlines progress made towards identifying agents capable silencing aberrant signalling through disruption complexation events.
