摘要: Summary Objective The pathogenesis of osteoarthritis (OA) includes cartilage degeneration, synovial inflammation, and bone changes. Slowly, the sequence inter-relationship these features is becoming clearer. Early models OA suggest thinning subchondral plate in addition to trabecular In present study changes were studied canine anterior cruciate ligament transection (ACLT)-meniscectomy model. This model characterized by intra-joint variability with respect damage (predominantly medial) loading (lateral unloading due a shifted axis). Methods 13 Labrador dogs, was induced removal medial meniscus. Twelve weeks later, integrity evaluated histologically using modified Mankin score (0–11), proteoglycan content determined Alcian Blue assay. Bone architecture tibia quantified micro-CT. Results Cartilage severe compartment (Mankin +3.5, glycosaminoglycan (GAG) −28%) mild lateral +1.6, GAG −15%). Thinning porosity only on side (−21%, +87%, respectively). Interestingly, structure did almost not occur (volume fraction −7%) but clear (−20%). Conclusion localized phenomenon related degeneration while are mechanical (un)loading. different mechanisms responsible for should be taken account when designing interpreting studies interfering turnover treatment OA.
