摘要: A number of independent insertions into bacteriophage lambda the translocatable tetracycline-resistance element Tn10 have been isolated and characterized. The physical positions relative orientations several such were determined. Two appear to lie in same orientation at or very near site cI gene, two more opposite position rex gene. Insertions genes cI, rex, cIII characterized genetically for their effects on expression nearby genes. appears exert a polar effect distal when it is inserted within an operon, even that operon under influence N-function. In addition, some way "upstream" cI. Lambda derivatives carrying give rise spontaneously occurring, tetracycline-sensitive deletions high frequencies. It likely formation these promoted by element. Lambda::Tn10 phages has undergone successive cycles translocation since its first isolation characterization analyzed. results confirm often retains functional integrity during many translocation. used generate chromosome Escherichia coli. This process recA function, seems be quite analogous Salmonella typhimurium as studied previously.
