摘要: We have identified a new putative transcription factor from the rat kidney, termed Kid-1 (for ischemia and developmentally regulated gene 1). belongs to C2H2 class of zinc finger genes. Its mRNA accumulates with age in postnatal renal development is detected predominantly kidney. levels decline after injury secondary or folic acid administration, two insults which result epithelial cell dedifferentiation, followed by regenerative hyperplasia differentiation. The low expression early development, when tissue recovering injury, suggests that product involved establishment differentiated phenotype and/or regulation proliferative response. deduced protein contains 13 fingers at COOH end groups 4 9 separated 32-amino-acid spacer. There are consensus sites for phosphorylation NH2 terminus non-zinc region as well spacer between 5. A shares extensive homology catalytic Raf kinases, feature shared only TFIIE among factors. To determine whether can modulate transcription, chimeric construct encoding (sense antisense) DNA-binding GAL4 was transfected into COS LLC-PK1 cells together chloramphenicol acetyltransferase (CAT) reporter plasmid containing binding sites, driven either minimal promoter simian virus 40 enhancer. CAT activity markedly inhibited sense compared antisense construct. our knowledge, this pattern developmental regulation, kidney expression, unique finger-containing proteins.
