Ophiopogon japonicus strains from different cultivation regions exhibit markedly different properties on cytotoxicity, pregnane X receptor activation and cytochrome P450 3A4 induction

摘要: Maidong, known as Ophiopogon japonicus, is one of the two basic ingredients Shenmai injection, which a widely used herbal preparation in traditional Chinese medicine (TCM) for treatment atherosclerotic coronary heart disease and viral myocarditis. Previously, ethanol extract Maidong activated pregnane X receptor (PXR) signaling pathway induced cytochrome P450 3A4 (CYP3A4) reporter gene raised concern herb-drug interactions (HDIs) when was combination with prescribed drugs metabolized by CYP3A4. Therefore, present study further investigated compared differences aqueous extracts (ee- ae-, respectively) strains, Zhe (ZM) Chuan (CM). Cytotoxicity, PXR activation CYP3A4 induction 3-(4,5)-dimethylthiahiazo-(-z-y1)-3,5-diphenytetrazoliumromide assay, assay reverse transcription-quantitative polymerase chain reaction analysis were examined. The observations showed that ee-ZM demonstrated significantly higher cytotoxicity, relatively weaker capability markedly stronger CYP3A4-inducing capacity than ee-CM. Compared to ae-CM, ae-ZM exhibited only slight or no difference on cytotoxicity induction, while significant lower level apparent. Collectively, from different producing areas possess properties upon drug-metabolizing enzyme inducing effect, attention should be paid selection strains planting regions into TCM preparations reducing potential adverse reactions HDIs.
