Nature of “Tau” immunoreactivity in normal myonuclei and inclusion body myositis

摘要: Sarcoplasmic accumulation of phosphorylated-tau has been widely stated to occur in and contribute the pathogenesis muscle disease inclusion body myositis. Twenty inflammatory myopathy 10 normal samples along with a range other tissues were stained anti-"tau" antibodies (tau-5, pS422, SMI-31). Myonuclear sarcoplasmic fractions prepared using differential solubilization laser-capture microdissection, immunoblots performed pS422 SMI-31 antibodies. All three demonstrated anti-tau immunoreactivity myonuclei from diseased muscle, but not nuclei tissues. Western blots showed against nuclear proteins outside region expected for phosphorylated-tau. Antibodies previously reported indicate abnormal IBM myofibers react recognize than tau. Normal contain neurofilament H or unidentified 200 kDa similar phosphorylated motifs accounting immunoreactivity.
