摘要: Rapid inactivation of metabolism is essential for accurately determining the concentrations metabolic intermediates in vivo state. We compared a broad spectrum energetic intermediate metabolites and neurotransmitters brains obtained by microwave irradiation to those freeze blowing, most rapid method extracting freezing rat brain. The many intermediates, cytosolic free NAD(P)+/NAD(P)H ratios, as well were not affected procedure. However, brain ATP about 30% lower, whereas ADP, AMP, GDP higher microwave-irradiated with freeze-blown brains. In addition, hydrolysis approximately 1 μmol/g ATP, major Mg2+-binding site, was related five-fold increase [Mg2+] (0.53 ± 0.07 mM blown vs. 2.91 mM ± 0.48 mM microwaved brains), determined from ratio [citrate]/[isocitrate]. Consequently, intracellular properties, such phosphorylation potential ∆G' significantly altered tissue. determinations some glycolytic TCA cycle metabolites, potential, do represent state when using microwave-fixed
