摘要: West Nile virus (WNV) is an emerging human pathogen for which specific antiviral therapy has not been developed. The therapeutic potential of RNA interference (RNAi) as a sequence-specific inhibitor WNV well demonstrated. Although multiple siRNA targets have identified within the genomic coding region, untranslated regions (UTR), encode cis-acting regulatory elements, remain relatively unknown. In and other flaviviruses, UTRs are located at termini. These form complex secondary structures, pose difficulty when designing effective targets. this study, we report identification in 3' UTR. were selected by predictor algorithms, synthesized short hairpin sequences from plasmid-based expression system. Vero cells stably expressing these had greatly diminished ability to support replication but related dengue virus, demonstrating that siRNAs suppressed viral manner. developed study could function therapeutics genetic tools investigate role UTR pathogenesis.
