Effect of the co-expression of methyltransferase activity on extracellular production of Taq I restriction endonuclease in recombinant E. coli cells

摘要: Taq I methylase gene was cloned and expressed constitutively under the control of tetracycline resistance promoter. The effect protection on growth kinetics, plasmid stability production MBP–Taq fusion protein by recombinant E. coli cells investigated both in shake flasks controlled bioreactor conditions. Shake flask experiments indicated that use nutritionally richer medium formulations may improve characteristics, also total extracellular endonuclease recovery yields protected cells. Under conditions, co-expression led to higher stabilities improved excretion levels considerably.
