摘要: The role of estrogen in the growth human breast cancers has been investigated at two levels. First, we have studied pS2 gene, whose transcription is stimulated by cancer cell line, MCF-7. gene product a small, secreted polypeptide currently unknown function, but with structural features similar to some factors. expression so far detected only MCF-7 cells and biopsies. Preliminary studies indicate that potential marker for hormone-dependent cancer. Ongoing will continue focus on implicated estrogen-mediated its possible use as estrogen-dependent tumors. Second, analyzed structure function ER. receptor stimulates interacting an ERE 5'-flanking region gene. A mutational analysis protein localized DNA-binding domain, which determines target specificity, hormone-binding domain. These domains appear be regions are absolutely required transcription-activating ER transfection assays reporter plasmids. N-terminal (regions B), necessary increasing efficiency using ERE, not vitellogenin may also play activation. Further progress characterization functional require genes more physiological chromatin environment, well detailed physical analyses structure.
