Effects of diacerein on cartilage and subchondral bone in early stages of osteoarthritis in a rabbit model

摘要: Background: Osteoarthritis is thought to be the most prevalent chronic and disabling joint disease in animals humans. At present, there no ideal treatment option. The aim of this study was assess effects with oral diacerein on articular cartilage, synovial membrane subchondral bone an experimental rabbit model osteoarthritis by micro-CT evaluation histological analysis. To purpose, surgically induced one knee 16 rabbits using contralateral as healthy controls. Treatment started three weeks later lasted eight weeks. Animals were divided into two groups for treatment: Placebo (treated daily saline) orally 1.5 mg/kg/day diacerein). Results: Sample analysis revealed that operated joint. placebo group showed a significant increase non-calcified cartilage thickness volume respect control important changes membrane; whereas parameters measured remained unchanged. In diacerein-treated results improvement OA all parameters, although not statistically significant. Conclusion: animal suggested may able ameliorate swelling surface alterations exert anti-inflammatory effect membrane, which might contribute improvement, well anabolic trabecular bone.
