摘要: Abstract Restoration of the Chol homeostasis in Central Nervous System (CNS) could be beneficial for treatment Huntington’s Disease (HD), a progressive, fatal, adult-onset, neurodegenerative disorder. Unfortunately, is unable to cross blood–brain barrier (BBB), thus novel strategy targeted delivery into brain highly desired. This article aims investigate production hybrid nanoparticles composed by and PLGA (MIX-NPs) modified with g7 ligand BBB crossing. We described impact ratio between components (Chol PLGA) formulation process (nanoprecipitation or single emulsion process) on physico-chemical structural characteristics, we tested MIX-NPs vitro using primary hippocampal cell cultures evaluating possible toxicity, uptake, ability influence excitatory synaptic receptors. Our results elucidated that both processes produce content higher 40%, meaning particle component active compound at same time. The impacted architecture reorganization leading some differences availability two types MIX-NPs. identified kinds are efficiently taken up neurons, able escape lysosomes release cells resulting an efficient modification expression receptors HD.
