Rapid Detection and Differentiation of Campylobacter jejuni, Campylobacter coli, and Campylobacter lari in Food, Using Multiplex Real-Time PCR

摘要: A multiplex real-time PCR assay based on four differently labeled TaqMan probes for detection and differentiation of the thermophilic Campylobacter species C. jejuni, coli, lari was established validated in food products. This combines two previously published assays jejuni coli with a newly developed an internal amplification control system. The selectivity method determined by analyzing 70 strains 43 other bacteria. sensitivity 50 fg DNA 500 per PCR. It possible to detect 1 10 CFU/25 g before preenrichment all three species. More than 400 samples various foods (poultry, seafood, meat) were analyzed after 48 h parallel conventional diagnostic culture biochemical identification. Using assay, 55.4% recognized as positive species, whereas only 40.3% positive. also detected contaminations different 32.6% poultry samples, finding epidemiological interest. Compared original method, which bacterial isolates this new detects distinguishes lari, analytical tool analysis, provides reliable extensive results within 2 days.
