摘要: Generation of antigen-specific humoral responses following vaccination or infection requires the maturation and function highly specialized immune cells in secondary lymphoid organs (SLO), such as lymph nodes tonsils. Factors that orchestrate dynamics these are still poorly understood. Currently, experimental approaches enable a detailed description system SLO have been mainly developed optimized animal models. Conversely, methodological humans based on use blood-associated material because challenging access to tissues. Indeed, only few studies were able provide discrete complex network cytokines, chemokines lymphocytes acting tissues after antigenic challenge. Furthermore, even fewer data currently available interaction occurring within micro-architecture SLO. This information is crucial order design particular strategies, especially for patients affected by chronic compromising medical conditions who under-vaccinated respond immunizations. Analysis different human high-throughput technologies, obtain ranging from gene signature protein expression cell phenotypes, needed dissect peculiarity each definite tissue. The main aim this review an in-depth current methodologies, proven evidence future perspectives analysis mechanisms immunization infections
