摘要: Publisher Summary This chapter discusses the cell-cycle synchronization, chromosome isolation, and flow sorting in plants. Plant cell populations synchronously traversing division cycle or accumulated within a specific phase of represent valuable model for studying wide range biological questions. Techniques sorting, production chromosome-specific libraries were initially developed mammalian species. The delay developing equivalent techniques cytometric analysis plant chromosomes was mainly due to difficulties obtaining suspensions that contained intact well-dispersed free contaminating cellular debris. procedure isolation involves four fundamental steps: (1) induction synchrony, (2) accumulation cells metaphase, (3) from cells, (4) chromosomes. For suspensions, problems posed by use tissues include high level mitotic variable degree stickiness clumping observed after treatment with drugs, tendency metaphase-blocked split into single chromatids, presence rigid wall which, comparison increases debris inhibits release during homogenization.
