摘要: How myosin interacts with actin to generate force is a subject of considerable controversy. The major debate centers on understanding at what point in generation the inorganic phosphate released respect lever arm swing, or powerstroke. Resolving controversy essential for how produced as well mechanisms underlying disease-causing mutations myosin. Recent structural insights into powerstroke have come from high-resolution structure previously unseen state and an electron cryomicroscopy (cryo-EM) 3D reconstruction actin–myosin–MgADP complex. Here, we argue that seemingly contradictory data time-resolved fluorescence resonance energy transfer (FRET) studies can be reconciled, put forward model actin.
