摘要: Complexes of RecA-DNA filaments, formed in the presence a non-hydrolyzable ATP analog, gamma S, aggregate together into regular bundles Mg2+. Electron micrographs several different forms RecA-double-stranded DNA have been analyzed: six supercoiled filaments at two concentrations Mg2+, and three single concentration The are all characterized by left-handed supercoiling component arising from non-integral number RecA subunits per turn helix these aggregates, about 6.15 units/turn. When single-stranded is used instead double-stranded DNA, aggregates composed many formed. These do not supercoil, consistent with symmetry close to 6.0 structures provided strong confirmation analysis isolated filaments. Since protomers within environments, they direct view conformations that may adopt same filament as result nonequivalent contacts. conformational changes we visualized quite large, apparent movements mass over distances greater than 2 nm. RecA-mediated strand exchange reaction highly dynamic process, which involves both unwinding stretching addition physical movement strands. It likely, therefore, seen represent states during its enzymatic activity.
