Differential modification of flavonoid and isoflavonoid biosynthesis with an antisense chalcone synthase construct in transgenic Lotus corniculatus

摘要: Three clonal genotypes of Lotus corniculatus L. (bird's foot trefoil) were transformed with an antisense chalcone synthase (CHS) gene construct made using a stress induced CHS17 cDNA from Phaseolus vulgaris under the control constitutive CaMV 35S promoter and Nos terminator via Agrobacterium rhizogenes. After initial screening, ten five co-transformation events each recipient genotype analysed. elicitation glutathione, level tannin accumulation was found to be increased in number root cultures derived low (S33) moderate (S50) genotypes. Six four lines S50 selected for more detailed study. The CHS integrated into genome, copy ranging 1 5 orientation confirmed by PCR. In cultures, transcript levels noted lines. Biochemical analyses glutathione-elicited-root indicated significant increase mean vestitol reduced. These results show that introduction heterologous resulted unpredicted molecular biochemical phenotype. Such findings are discussed relation manipulation this complex multigene family.
