摘要: The proper staining of the plasma membrane (PM) is critical in bioimaging as it delimits cell. Herein, we developed MemBright: a family six cyanine-based fluorescent turn-on PM probes that emit from orange to near-infrared when reaching PM, and enable homogeneous selective with excellent contrast mono two-photon microscopy. These are compatible long-term live cell imaging immunostaining. Moreover, MemBright label neurons brighter manner than surrounding cells allowing identification acute brain tissue section neuromuscular-junctions without any use transfection or transgenic animals. At last, were used super-resolution unravel dendritic spines neck. 3D multicolor dSTORM combination immunostaining revealed en-passant synapse displaying endogenous glutamate receptors clustered at axonal-dendritic contact site. thus constitute universal toolkit for biology neuroscience biomembrane variety microscopy techniques.
