摘要: Abstract Omeprazole, a widely used antiulcer drug, has been enantiomerically separated at semipreparative scale on polysaccharide based chiral stationary phase by supercritical fluid chromatography (SFC). For this work, modular chromatograph was adapted to operate and Chiralpak AD (250 mm × 10 mm) column used. The effect of two organic modifiers (ethanol 2-propanol) studied, different injection volumes concentrations the omeprazole racemic mixture were evaluated in order obtain high enantiomeric purities production rates. Better results achieved using concentration overloading instead volume overloading. recoveries decreased when requirements purity or load increased, but it possible recover 100% both enantiomers an higher than 99.9% under some loading conditions, like injecting 1 2 ml solution 3 g/l. As far as rates are concerned, best result for S-(−)-omeprazole that (27.2 mg/h) with sample 10 g/l 2 ml, while 4 ml better case R-(+)-omeprazole (20.5 mg/h).
