摘要: The coiled-coil domain is a structural motif found in proteins that participate variety of biological processes. Aberrant expression such has been shown to be associated with the malignant behavior human cancers. In this study, we investigated role specific family member, containing 109B (CCDC109B), gliomas. We confirmed CCDC109B was highly expressed high grade gliomas (HGG; WHO III–IV) using immunofluorescence, western blot analysis, immunohistochemistry (IHC) and open databases. Through Cox regression analysis Cancer Genome Atlas (TCGA) database, levels were inversely correlated patient overall survival it could serve as prognostic marker. Then, serious cell functional assays performed glioma lines, U87MG U251, which indicated silencing attenuated proliferation migration/invasion both vitro vivo. Notably, IHC staining primary samples interestingly revealed localization elevated necrotic areas are typically hypoxic. Moreover, small interfering RNA (siRNA) inhibiters HIF1α led decreased Transwell assay further showed critical factor mediating HIF1α-induced migration invasion. Our study elucidated for an oncogene marker may provide novel therapeutic target treatment glioma.
