Rac-ing to the plasma membrane: the long and complex work commute of Rac1 during cell signaling.

摘要: The functional cycle of the Rac1 GTPase involves a large number steps, including post-translational processing, cytosolic sequestration by RhoGDIs, translocation to specific subcellular localizations, activation GDP/GTP exchange, inactivation GTP hydrolysis, and re-formation Rac1/RhoGDI inhibitory complexes. Here, we summarize current knowledge about regulation those steps. In addition, discuss recently described, cytoskeletal-dependent feed-back loop that favors efficient Rac subfamily proteins during cell signaling. This route is mediated heteromolecular protein complex composed cytoskeletal coronin1A, Dbl family member ArhGEF7, serine/threonine kinase Pak1, dimer. promotes F-actin-rich juxtamembrane areas, Pak1-dependent release from complex, activation. pathway important for optimal signaling EGF receptor, integrins, antigenic T-cell receptor.
