摘要: BACKGROUND Valvular disease is characterized in part by lipid deposition, but systematic analysis of the patterns global expression healthy and diseased valve tissues are unknown. This due to tissue limitations for lipidomic preparations technologies evaluating distribution tissues. The study aim was examine application matrixassisted laser desorption ionization imaging mass spectrometry (MALDI IMS) aortic during development disease, as an approach detect map lipids ultimately better understand structure function. METHODS Established MALDI IMS strategies were applied thin sections heart valves corresponding morphological features. Healthy prenatal adult ovine evaluated using developed techniques. Lipid levels compared between Wilcoxon rank sum testing area under receiver operating curves. A classification algorithm used determine distinct signatures extracellular matrix (ECM) substructures, including fibrosa spongiosa layers. examined from pediatric patients with congenital stenosis (CAVS). RESULTS decreased when valves. Classification algorithms reported mapping ECM substructures valve, could not distinguish amorphous structures at pre-natal day 5. In CAVS, in-situ aggregation species showed unique patterning both concurrent divergent disarray. Fatty acid content varied normal human CONCLUSIONS provides a new useful evaluate biology tissue. These findings define role regulation demonstrate deregulation disease.
