Identification of enteroviruses by indirect immunofluorescence using monoclonal antibodies

摘要: Abstract Background: Serum neutralization (Nt) is used most often to type enterovirus isolates, but it labor-intensive, expensive, and supplies of reference antisera for Nt are limited. Alternative methods typing needed. Objectives: To investigate the use indirect immunofluorescence (IFA) with commercially available monoclonal antibodies (MAbs) as an alternative identification enteroviruses. Study design: Two MAb blends (one coxsackie B viruses one echoviruses 4, 6, 9, 11, 30, 34) a A9 were screen 465 clinical isolates over period two years. Virus which tested positive typed individual MAbs respective blend. Individual polioviruses 1, 2, 3 acquired late in study 45 viral isolates. Results: The identified 251 (54%) total number tested. IFA results 451 agreement conventional methods. sensitivity using was 93% specificity 99%. Thirteen discrepant negative by IFA, twelve echovirus 30 isolate A9. remaining both B5, only. Conclusions: highly specific enteroviruses, may not be sensitive enough identify all strains within type. Procedures utilize confirm final decrease turnaround time reduce cell culture tubes required each isolate.
