Synergy Maps: exploring compound combinations using network-based visualization

摘要: The phenomenon of super-additivity biological response to compounds applied jointly, termed synergy, has the potential provide many therapeutic benefits. Therefore, high throughput screening compound combinations recently received a great deal attention. Large libraries and feasibility all-pairs can easily generate large, information-rich datasets. Previously, these datasets have been visualized using either heat-map or network approach—however visualizations only partially represent information encoded in dataset. A new visualization technique for pairwise combination data, “Synergy Maps”, is presented. In Synergy Map, about synergistic interactions integrated with their properties (chemical structure, physicochemical properties, bioactivity profiles) produce single visualization. As result relationships between may be investigated simultaneously, thus afford insight into synergy observed screen. An interactive web app implementation, available at http://richlewis42.github.io/synergy-maps , developed public use, which find use navigating filtering larger scale This tool recent dataset anti-malarials, tested against Plasmodium falciparum, preliminary analysis given as an example, illustrating disproportionate synergism histone deacetylase inhibitors previously described literature, well suggesting hypotheses future investigation.
