摘要: OBJECTIVE: To investigate the effect of endothelin receptor A (ETA) and B (ETB) blockade on liver microcirculation oxygenation during intra-abdominal pressure (IAP) increase. METHODS: Fifteen anesthetized pigs were randomized to receive either nonselective endothelin-1 (ET-1) blocker tezosentan (TG, n = 7) or saline (CG, n = 8). Helium was insufflated increase IAP from 0 25 mmHg. Stroke volume variability maintained ≤ 12% with colloid infusion. Total blood flow (TLBF), hepatic microcirculatory (MBF), tissue (HpO2), hyaluronic acid plasma disappearance rate (PDR) indocyanine green (ICG) recorded. RESULTS: TLBF remained mostly unaltered in both groups at low decreased only CG As increased, a significant decline MBF observed without correlation cardiac output TLBF. In CG, HpO2 as early 10 mmHg high levels pressure. However, TG decrease PDR ICG 25 mmHg (P = 0.046 P = 0.009 respectively). These changes correlated (r = 0.793). CONCLUSION: Blocking ETA ETB receptors partially protects sinusoidal circulation against stress induced by IAP.
