The sperm motility pattern in ecotoxicological tests. The CRYO-Ecotest as a case study.

摘要: Changes in environmental stressors inevitably lead to an increasing need for innovative and more flexible monitoring tools. The aim of this work has been the characterization motility pattern cryopreserved sea bream semen after exposure a dumpsite leachate sample, identification best representative parameters be used as endpoints ecotoxicological bioassay. Sperm evaluated either by visual computer-assisted analysis; concerning on activation those describing it times (duration parameters) have assessed, discerning them terms sensitivity, reliability methodology assessment means multivariate analyses. EC50 values ranged between 2.3 4.5ml/L, except total motile percentage (aTM, 7.0ml/L), which proved less sensitive among all tested parameters. According analyses, difference sensitivity "activation" respect "duration" ones can inferred; contrary, seem equally informative sperm or rapid sub-population, well seems not discriminating. In conclusion, CRYO-Ecotest is multi-endpoint bioassay that considered promising tool, characterized high plasticity, its easy tailored each time according different needs quality programs.
