摘要: Reverse genetics approaches are critical for uncovering complex biological processes and genetic engineering. Clustered regularly interspaced short palindromic repeats (CRISPR)–CRISPR-associated (Cas) system (CRISPR/Cas9) has been widely used generating mutants in a large number of species. The Nile tilapia (Oreochromis niloticus), gonochoristic teleost with XX/XY sex determination system, is good model studying fish reproduction. Here, we describe detailed procedure gene mutation by CRISPR/Cas9. This protocol covers selection target sites, vitro RNA transcription, artificial insemination microinjection one cell stage embryos. We also provide details detection somatic germline transmitted mutations, fast establishment mutant lines discuss some practical advices. will facilitate broader applications CRISPR/Cas9 studies as well other aquaculture fishes.
