Isolation of Functional Subsets of Human Peripheral Blood Monocytes

摘要: Monocytes were isolated by counterflow centrifugation of Ficoll-Hypaque separated peripheral blood mononuclear cells. The monocytes formed a bimodal volume distribution “large” and “small” phagocytic esterase-positive, peroxidase-positive cells with peaks at 470 410 µ3, respectively. large predominately Fc receptor positive, able to lyse both sensitized human chicken erythrocyte targets in ADCC assays, whereas the small largely FcR negative inactive against targets. However, was seen some fractions containing probably due FcR+ lymphocytes (K cells) those fractions. These experiments establish that are effectors monocyte is heterogeneous size, function, surface distribution.
