摘要: The pharmacokinetics of propofol in a dose 2.5 mg kg−1 given via vein the antecubital fossa were studied 18 patients. Anaesthesia was maintained with nitrous oxide oxygen all effects pretreatment fentanyl (n = 6) and maintenance halothane on also investigated. Pretreatment resulted prolonged apnoea four No serious side occurred. unpretreated patients who only can be described by three-compartment open mammalian model very rapid distribution (T½β about 3 min), elimination 45 min) slower final phase (T½γ 300 min). total body clearance (1.91 litre min−1). Propofol initially distributed into relatively large central compartment (41.3 litre) extensively redistributed (V55 305 litre; Vγ 722 litre). Throughout sampling period mean blood concentrations for pretreated 50% higher than only. Mean intermediate between those other two groups.
