摘要: Morphinone reductase, produced by Pseudomonas putida M10, catalyses the NADH-dependent saturation of carbon-carbon double bond morphinone and codeinone, is believed to be involved in metabolism morphine codeine. The structural gene encoding designated morB, was cloned from Ps. M10 genomic DNA use degenerate oligonucleotide probes based on elements amino acid sequence purified enzyme. Sequence analysis characteristics indicated that reductase related flavoprotein alpha/beta-barrel oxidoreductases, particularly similar Old Yellow Enzyme Saccharomyces spp. oestrogen-binding protein Candida albicans. Expressed tags several plant species show high homology these enzymes, suggesting presence a family enzymes conserved plants fungi. Although bacterial proteins are known, appears more eukaryotic proteins. overexpressed Escherichia coli, has potential applications for industrial preparation semisynthetic opiates.
