摘要: Colorectal cancer (CRC) is currently the third leading cause of cancer-related mortality in world. U.S. Food and Drug Administration-approved circulating tumor markers, including carcinoembryonic antigen, carbohydrate antigen (CA) 19-9 CA125 were used as prognostic biomarkers CRC that attributed to low sensitivity diagnosis CRC. Therefore, our purpose develop a novel strategy for clinical early diagnosis. We mass spectrometry (MS) methods such nanoLC-MS/MS, targeted LC-MS/MS, stable isotope-labeled multiple reaction monitoring (MRM) MS coupled test machine learning algorithms logistic regression analyze plasma samples from patients with early-stage CRC, late-stage healthy controls (HCs). On basis methods, 356 peptides identified, 6 differential expressed verified, finally three corresponding wheat germ agglutinin (WGA)-captured proteins semi-quantitated 286 (80 HCs 206 CRCs). The peptide combination PF454-62, ITIH4429-438, APOE198-207 achieved 84.5%, specificity 97.5% an AUC 0.96 In conclusion, study demonstrated WGA-captured levels may serve highly effective diagnostic
