摘要: Streptomyces albus J1074 is one of the most popular and convenient hosts for heterologous expression gene clusters directing biosynthesis various natural metabolic products, such as antibiotics. This fuels interest in elucidation genetic mechanisms that may limit secondary metabolism J1074. Here, we report generation initial study mutant, deficient bldA tRNALeuUAA, only tRNA capable decoding rare leucyl TTA codon Streptomyces. The deletion resulted a highly conditional Bld phenotype, with depleted formation aerial hyphae on certain solid media. In addition, mutant was unable to produce endogenous antibacterial compounds two antibiotics, moenomycin aranciamycin, whose directed by TTA-containing genes. We have employed new codon-specific β-galactosidase reporter system provide evidence impaired translation TTA. discussed possible reasons differences phenotypes mutants described here previous studies, providing knowledge bldA-based regulation antibiotic biosynthesis.
